Research Overview
Why two mechanisms are combined
Mitochondrial capacity can be changed in two ways: build more respiratory machinery, or run existing machinery less efficiently so more substrate is consumed per unit of ATP produced. SLU-PP-332 addresses the first through ERR-driven transcription; BAM-15 addresses the second through proton leak. Combining them in one research article lets investigators ask whether the two effects are additive, redundant, or whether raising leak blunts the transcriptional signal.
The ERR arm
The uncoupler arm
- Oxygen consumption rate and proton leak measured by extracellular flux analysis
- Mitochondrial membrane potential assessed with potentiometric dyes
- Selectivity checks confirming plasma-membrane potential is not comparably affected
- Substrate oxidation and heat production in isolated mitochondria
