Cagrilintide and the amylin axis

Amylin is co-secreted with insulin from pancreatic beta cells and signals through amylin receptors — heterodimers of the calcitonin receptor with receptor activity-modifying proteins. Native amylin aggregates readily, which limited its research use; cagrilintide is an engineered analogue with substitutions and lipidation intended to give a soluble, long-acting molecule. Published work has examined its receptor selectivity profile and its behaviour in models of food intake and body-weight regulation.

Why the two pathways are studied together

Large lipidated peptides are sensitive to shear, so handling matters more here than with short sequences. Let the vial reach room temperature, sanitise the stopper, then run bacteriostatic water slowly down the vial wall and let the cake dissolve with gentle swirling. Never shake or vortex — foaming at the air-liquid interface is the main route to loss of intact peptide. Expect a clear, colourless solution. Store reconstituted material at 2–8 °C, protected from light, aliquot rather than freeze-thawing repeatedly, and record the reconstitution date on the vial.
