Why glutathione is a standard redox reagent
Cells maintain millimolar cytosolic GSH, and the ratio of reduced GSH to oxidised GSSG is widely used as a quantitative index of redox state. Glutathione peroxidases use GSH to reduce hydrogen peroxide and lipid hydroperoxides; glutathione S-transferases conjugate it to electrophiles for export; glutaredoxins use it to reverse protein S-glutathionylation. Because those reactions consume GSH stoichiometrically, its concentration behaves as a dynamic variable rather than a fixed background quantity.

Intranasal and mucosal delivery research
The nasal epithelium is thin, richly perfused and adjacent to olfactory and trigeminal pathways, which is why mucosal routes are studied for molecules that are poorly absorbed orally. Glutathione is a polar, ionisable tripeptide, and orally administered GSH is extensively hydrolysed by gamma-glutamyl transpeptidase at the intestinal brush border — a well-documented reason researchers compare non-oral formats. Whether nasal presentation produces meaningful uptake is compound-specific and is itself the subject of investigation, so route-comparison designs normally include a parenteral reference arm.
Formulation and stability considerations
Thiols oxidise in solution. Oxygen exposure, elevated pH, warmth and trace transition metals all accelerate GSH conversion to GSSG, so solution-format work is generally done cold, in the dark and within a defined in-use window. Investigators comparing lots often verify free-thiol content directly rather than assuming the labelled value persists over time.
