Local versus systemic exposure

An open question in the literature is how much of an orally delivered quantity reaches systemic circulation intact versus acting locally on gut epithelium. Papers that report systemic effects after oral administration in rodents exist, but pharmacokinetic characterisation in humans is essentially absent, so the distinction between local and systemic action is not resolved.

Practical advantages of a capsule

Fixed unit content removes weighing and reconstitution error from a study design. In multi-time-point or multi-animal work, that consistency between units is worth more than the flexibility of a vial, since the main preparation variable has been eliminated before the experiment starts.

Evidence limits

Keep the bottle closed, dry and away from direct light and heat; refrigeration is not required but does extend shelf stability, and the desiccant supplied with the bottle should stay inside it. Peptide powders are hygroscopic, so avoid leaving the container open in humid conditions and do not transfer capsules to unsealed containers for storage. Record the lot number from the label against the matching certificate of analysis at the start of a study so unit content can be traced afterwards. Do not use capsules whose shells have softened, discoloured or fused.
