An open question in the literature is how much of an orally delivered quantity reaches systemic circulation intact versus acting locally on gut epithelium. Papers that report systemic effects after oral administration in rodents exist, but pharmacokinetic characterisation in humans is essentially absent, so the distinction between local and systemic action is not resolved.
Practical advantages of a capsule
Fixed unit content removes weighing and reconstitution error from a study design. In multi-time-point or multi-animal work, that consistency between units is worth more than the flexibility of a vial, since the main preparation variable has been eliminated before the experiment starts.
