Research Overview
Why an oral GLP-1 tablet is difficult
The role of the absorption enhancer
- SNAC raises local pH in the gastric microenvironment, reducing pepsin-mediated degradation
- It promotes transient, reversible permeation across the epithelium in the immediate vicinity of the tablet
- Because the effect is local and short-lived, the enhancer-to-peptide ratio and the site of tablet erosion strongly influence measured uptake
- Co-administered fluid volume and fasting state have been reported to shift absorption substantially
What laboratories study with this format
Typical endpoints include dissolution and disintegration profiles under different media, stability of the peptide within the tablet matrix over time, permeation across Caco-2 or ex vivo intestinal tissue models, and comparative pharmacokinetics against a parenteral reference. Variability between subjects and between administrations is itself a frequent object of study.
