Chemistry and origin
Andarine belongs to the aryl-propionamide class of SARMs developed from non-steroidal antiandrogen chemistry at the University of Tennessee and later at GTx. Substituting the B-ring of the bicalutamide-type scaffold shifted the molecules from receptor blockade toward partial agonism at the androgen receptor while retaining oral exposure and avoiding the 5-alpha-reduction and aromatisation pathways that apply to steroidal androgens.
Reported effects and limits
- Andarine acts as a partial agonist, so studies note it can antagonise stronger androgens in some tissue contexts.
- Early human work reported reversible visual disturbance, described as a yellowish tint and altered dark adaptation, attributed to the compound's structural relationship to a class of chromophore-affecting molecules.
