What is the difference between PACAP 38 and PACAP 27?
Both come from the same precursor and share an identical N-terminal 27 residues; PACAP 38 carries an additional C-terminal extension. PACAP 38 predominates in most mammalian tissues, and the two forms can differ in receptor selectivity and stability in experimental systems.
Which receptors does PACAP act on?
Published work describes three class B G protein-coupled receptors: PAC1, which prefers PACAP over VIP, and VPAC1 and VPAC2, which bind both peptides. Coupling is mainly to adenylate cyclase, with additional phospholipase C signalling for some PAC1 variants.
Are non-mammalian PACAP sequences available?
Yes. PACAP is highly conserved, and human, rat, mouse and ovine sequences are often identical over the mature peptide, while frog and other non-mammalian forms are listed separately for comparative and evolutionary studies of the VIP/secretin superfamily.
