GHRP-2 as a refinement of the class
The GHRP series began with GHRP-6, a hexapeptide that released growth hormone through a receptor distinct from the GHRH receptor. GHRP-2 followed, incorporating D-amino acids and a 2-naphthylalanine residue that raise potency and stability. In comparative pharmacology it is generally described as releasing more growth hormone per unit mass than GHRP-6 while producing far less appetite stimulation, which is why it displaced the original in study designs where feeding behaviour would be a confound.
Why the combination is studied
- GHRP-2 and the GHRH analogue engage separate receptors expressed on the same pituitary cell.
- GHS-receptor activation suppresses somatostatin tone, which otherwise limits the amplitude of a GHRH response.
- Animal and pituitary-cell studies report a larger measured growth hormone pulse from the pair than from either compound alone.
- Both components are short-acting, so the resulting stimulus is pulsatile rather than sustained — relevant in a feedback-regulated axis.
