Subcutaneous (abbreviated SC, SQ or subQ) describes delivery into the loose adipose and connective tissue layer beneath the dermis, above muscle. In published peptide work it is the most common parenteral route in rodent and large-animal models, and it is the route used for the approved peptide drugs that research-grade analogs are modelled on.

Why the subcutaneous route matters in peptide research

The tissue is poorly vascularised relative to muscle, so absorption is slower and flatter than intravenous or intramuscular delivery: the injected volume forms a depot that empties over minutes to hours. Molecules below roughly 16 kDa are reported to enter blood capillaries directly, while larger ones are taken up preferentially by lymphatics, which adds a further delay. That gives a lower peak concentration and a longer measurable tail, and it is one reason bioavailability by this route is usually high but rarely complete.

Volume is the practical constraint. Small aqueous volumes are the norm, which pushes concentration decisions back into the reconstitution calculation and makes U-100 syringe graduations and needle gauge selection part of study planning.
