Acetate salt means the peptide’s basic residues are paired with acetate as the counter-ion rather than the trifluoroacetate left behind by synthesis. Because acetate is not generated by the SPPS process, it has to be installed deliberately — typically by repeated lyophilisation from dilute acetic acid or by ion-exchange chromatography.
Why acetate form matters in peptide research
Acetate is a normal metabolic intermediate and is biologically innocuous at the levels involved, whereas residual TFA is reported to affect viability and proliferation in some cell lines. That is why approved peptide products are almost invariably acetates — leuprolide acetate, octreotide acetate, larazotide acetate — and why cell-based, immunological and in-vivo research frequently specifies acetate form.
Two practicalities. Exchange is not free: it adds cost, causes some material loss, and is never quite complete, so a residual TFA figure should still be reported. And acetate has its own mass, roughly 60 Da per equivalent, so gross vial mass again exceeds net peptide content and a molarity calculation must use the figure on the certificate of analysis.
