An excipient is any substance in a formulation other than the active peptide. In a lyophilised research vial the candidates are bulking agents such as mannitol, cryoprotectants such as sucrose or trehalose, buffer salts, tonicity agents and, in solutions, preservatives such as benzyl alcohol.
Why excipients matter in peptide research
They change three things researchers actually observe. Appearance first: a few milligrams of peptide alone may leave only a thin film on the glass, whereas the same mass with a bulking agent forms a visible cake — so cake size says more about formulation than about peptide content. Second, behaviour on reconstitution, since buffering excipients set the pH the peptide meets and can move it away from its isoelectric point. Third, mass accounting: labelled vial mass may or may not include excipient weight, which matters directly for any molarity calculation.
Excipients also interfere downstream. Buffer salts and sugars can affect assay readouts, and preservatives are incompatible with some cell work.
