NMN (nicotinamide mononucleotide) is the immediate biosynthetic precursor of NAD+ in the mammalian salvage pathway. NAMPT converts nicotinamide plus PRPP into NMN; NMNAT enzymes then adenylylate NMN to NAD+ in one further step. Because NAMPT is the rate-limiting enzyme of that pathway, supplying NMN downstream of it is the logic behind its use in metabolic experiments.
Why NMN matters in research
Rodent work published from about 2011 onward reported raised tissue NAD+ and changes in insulin sensitivity, mitochondrial and vascular readouts after NMN administration; a small number of human trials since 2020 have reported biomarker changes with modest and inconsistent functional endpoints. A live mechanistic argument concerns transport: NMN is phosphorylated and polar, and some data indicate it is dephosphorylated to nicotinamide riboside at the cell membrane before uptake, while other work supports a dedicated transporter (Slc12a8). That dispute is why NMN, NR and NAD+ itself are frequently compared head-to-head rather than regarded as interchangeable.
Regulatory note: US regulatory classification of NMN as a dietary-supplement ingredient has been contested since 2022, which is one reason it is supplied here as research-grade material only.
