Stromal Cell-Derived Factor (SDF-1 / CXCL12) — frequently asked

What is SDF-1?
SDF-1, also called CXCL12, is a constitutively expressed CXC chemokine produced by stromal cells. SDF-1 alpha is the most studied splice variant and adopts the standard chemokine fold with a flexible receptor-activating N-terminus.
Which receptors does CXCL12 use?
It is reported to signal mainly through CXCR4, a G protein-coupled receptor, and also to bind ACKR3, formerly CXCR7, which is generally described as a scavenging or arrestin-biased receptor rather than a classical signalling one.
How is SDF-1 used in the laboratory?
Common uses include chemotaxis and migration assays, calcium flux and receptor internalisation experiments, and stem or progenitor cell culture work. It also serves as a reference standard in chemokine immunoassay development.
