Why is normal rabbit serum needed?
Primary antibody is present in trace amounts in a competitive assay. Normal rabbit serum supplies additional carrier immunoglobulin so that the second antibody forms a precipitate large enough to separate bound tracer from free tracer reliably.
What does the second antibody do?
The goat anti-rabbit antibody recognises the rabbit primary antibody and precipitates the resulting immune complexes. This provides the bound-versus-free separation step that must occur before the radioactivity in each fraction is counted.
