What is an epitope tag peptide used for?
It is the free synthetic version of a tag sequence fused to a recombinant protein. Because it competes for the same antibody or resin binding site, it is used to elute tagged protein gently and to block antibody binding in specificity controls.
Why elute with peptide instead of low pH?
Competitive peptide elution releases the tagged protein under near-neutral, non-denaturing conditions. This is generally preferred when enzymatic activity, folding or an intact multi-protein complex needs to survive the purification.
How does hexahistidine differ from the antibody-recognised tags?
Hexahistidine binds immobilised metal ions such as nickel or cobalt rather than an antibody, so it is normally eluted with imidazole or a pH change. FLAG, HA and Myc tags are recognised by specific antibodies and can be competed off with the corresponding free peptide.
