Presenilin-1 is a multipass membrane protein that forms the catalytic core of the gamma-secretase complex, alongside nicastrin, APH-1 and PEN-2. It is itself cleaved into N-terminal and C-terminal fragments that remain associated within the active enzyme.

Gamma-secretase cleaves amyloid precursor protein to generate amyloid-beta peptides and processes Notch to release its signalling domain. Presenilin-1 mutations are a leading described cause of early-onset familial Alzheimer disease, making the protein central to neurodegeneration research.
