What is osteopontin?
Osteopontin is an acidic, heavily modified secreted phosphoprotein encoded by the SPP1 gene. It is intrinsically disordered rather than compactly folded, and it presents short linear motifs that mediate binding to integrins, CD44 and mineral surfaces.
Why is proteolytic processing important?
Cleavage by thrombin and by matrix metalloproteinases is reported to expose additional integrin-binding motifs that are not accessible in the intact protein. Fragments spanning that region are therefore studied separately to examine motif-specific adhesion effects.
How are these peptide fragments used?
They serve as immunogens and blocking peptides for antibody work, as calibration standards in immunoassays, and as defined sequences in cell adhesion and migration assays where a single motif is to be tested without the rest of the protein.
