Opiorphin — frequently asked

What is opiorphin?
Opiorphin is the human salivary pentapeptide Gln-Arg-Phe-Ser-Arg, reported to be released from proline-rich protein 1. It is considered the human counterpart of the rat peptide sialorphin and is classed as an endogenous ectopeptidase inhibitor.
How does opiorphin act?
Published work describes inhibition of neprilysin and aminopeptidase N, the enzymes that degrade enkephalins. The peptide is therefore reported to act indirectly, by prolonging the local availability of endogenous opioid peptides rather than binding opioid receptors itself.
Why are modified analogs listed?
The pGlu-modified form reflects a cyclisation that N-terminal glutamine can undergo, while the Tyr-extended version adds a residue suitable for radioiodination. Both let researchers study stability, detection and tracer applications alongside the native sequence.
