What are neurotrophins?
Neurotrophins are a family of secreted dimeric growth factors comprising nerve growth factor, brain-derived neurotrophic factor, neurotrophin-3 and neurotrophin-4. They share a cystine knot structure and are generated by cleavage of larger precursor proteins.
How do neurotrophin-3 and neurotrophin-4 differ?
Neurotrophin-3 is generally described as preferring TrkC while also showing some activity at other Trk receptors, whereas neurotrophin-4 is associated mainly with TrkB. Both additionally bind the p75 neurotrophin receptor shared across the family.
How are recombinant neurotrophins used?
They are commonly added to primary neuronal or cell line cultures to support survival and differentiation, and used as stimuli in receptor phosphorylation assays. Iodinated versions serve as tracers in quantitative receptor binding experiments.
