MHC Epitope & Antigen Peptides — frequently asked

What is an MHC epitope peptide?
It is a short synthetic peptide matching the fragment that an MHC molecule presents on the cell surface to a T cell. Class I epitopes are usually 8 to 10 residues, class II epitopes commonly 13 to 20, and the sequence is what determines which T-cell receptor recognises it.
Why do researchers use defined epitopes instead of whole antigen?
A single defined sequence removes the processing step, so a response can be attributed to one epitope rather than an unknown mixture. This makes epitopes standard reagents for tetramer staining, ELISpot readouts and quantifying antigen-specific T-cell frequencies.
Does the MHC restriction matter when selecting a peptide?
Yes. Each epitope is reported for a particular restriction element, such as a human HLA allele or a mouse H-2 haplotype. A peptide will generally not stimulate a response in cells that do not carry the matching allele, so the published restriction should be checked before use.
