What are enkephalins?
Enkephalins are endogenous opioid pentapeptides, principally Met-enkephalin and Leu-enkephalin, released from the proenkephalin A precursor. Their conserved N-terminal Tyr-Gly-Gly-Phe sequence is reported to drive recognition at opioid receptors, with delta and mu subtypes most commonly implicated.
Why are analogues such as DAMGO used?
Native enkephalins are degraded quickly by aminopeptidase and neprilysin activity, which complicates assays. Stabilised analogues including DAMGO, DPDPE and [D-Ala2, D-Leu5]-enkephalin resist cleavage and show clearer receptor selectivity, making them reliable reference tools in receptor pharmacology.
Which species variants exist?
The enkephalin pentapeptides themselves are identical across mammals, so those products carry no species label. Larger proenkephalin-derived fragments such as amidorphin and peptide F differ between species and are catalogued as bovine, human, rat or porcine sequences.
